超声辅助VEGF165转染骨髓间充质干细胞的实验研究

摘要 目的:探讨骨髓间充质干细胞定向分化血管内皮细胞的可行性研究。方法:采用超声辅助电穿法,将VEGF165基因的质粒PEGFP-C1转染至骨髓间充质干细胞,转染后于荧光显微镜下观察绿色荧光蛋白(GFP)表达情况。结果:超声辅助转染5h后可见细胞内有GFP表达,10h后可达高峰。结论:VEGF基因转染骨髓间充质干细胞后,骨髓间充质干细胞内有GFP表达,提示细胞转染成功,骨髓间充质干细胞定向分化为血管内皮细胞具有可行性。 Objective:To study the feasibility of medulla MSCs differentiation directionally to endothelial cells. Method:Observe the expression of green fluorescence protein(GFP) in medulla MSCs after transfection of VEGF165 vector,PEGFP-C1 by ultrasound under the fluorescence microscope.Result:GFP expression was seen 5 hours after transfection,and reached the peak 10 hours later.Conclusion: It is possible that medulla MSCs could be differentiate directionally to endothelial cells after the transfection of VEGF vector.
