摘要
目的探讨正常状态下不同胎龄大鼠神经干细胞(neuralstemeells,NSCs)在大脑中的分布。方法将孕鼠随机分为胎龄11天组、15天组和19天组,用免疫组织化学方法检测其大脑神经干细胞中的巢蛋白(Nestin),经图像分析比较其差异。结果胎龄11天组Nestin阳性细胞分布于整个神经管上皮层,随着胎龄的增加,Nestin阳性细胞主要在室管膜上皮层,细胞数逐渐增加,并向周围迁移;结论大鼠在正常胚胎发育过程中,随着胎龄的增加,其脑室室管膜及室管膜下区神经干细胞逐渐增多。
Objective To study the contribution of neural stem cells(NSCs)in rats from diffent embryonic days. Methods 12 female Wistar rats were divided randomly into three groups: embryonic days 11 group(E11) 、 (E15) and E19. The expression of Nestin in brain of embryos was studied with immunohistochmistry and the quantity of postive cells were analyzed by image processing system. Result In E11,the Nestin-postive cells were contributed throughout neural tube. With the increasing of the embryonic days, they aggregated mainly in ependymal and subependymal zone, and the number of Nestin-postive cells increased gradually. Conclusion With the increasing of the embryonic days, the NSCs in brain increase gradually in the proceeding of the normal development of embryo of rat.
