摘要
目的:观察针刺治疗缺血缺氧性脑性瘫痪幼鼠脑组织含水量和细胞凋亡情况,探讨其对促进神经功能恢复的影响。方法:实验于2004-09/2005-02商丘医学高等专科学校生理学教研室神经生物学实验室进行。7d龄新生Wistar大鼠62只,随机分为3组。①模型+针刺组30只,制作缺血缺氧性脑瘫幼鼠模型后12h,采用WQ-10DI型多用电子穴位治疗仪针刺百会,曲池,内关,涌泉穴位,固定频率5Hz,连续波,持续刺激20min。间隔24h后重复进行,连续7d。②模型组20只,制作缺血缺氧性脑瘫幼鼠模型,不做针刺治疗。③正常对照组12只,不做处理。各组动物于处理后7d处死,检测其脑组织含水量[(湿重-干重)/湿重×100%]及脑细胞凋亡情况(采用HPIAS-1000型图像分析系统对原位切口末端标记切片进行扫描,分析各组吸光情况,吸光度越高,说明表达越强)。结果:62只幼鼠均进入结果分析。①脑组织含水量测定结果:正常对照组平均为(79.3±2.5)%。模型+针刺组明显低于模型组[(82.2±1.9),(85.6±2.7)%,F=9.32,P<0.05]。②细胞凋亡检测结果:正常对照组鼠脑内基本没有凋亡细胞。模型+针刺组吸光度明显低于模型组吸光度(0.203±0.065,0.254±0.071,t=2.916,P<0.05)。结论:针刺可以通过减轻脑组织水肿和减少脑细胞凋亡两方面来促进缺血缺氧性脑瘫幼鼠神经功能的恢复。
AIM:To observe the effect of acupuncture on cell apoptosis and water content in brain tissue in neonatal rats with hypoxiaischemia cerebral palsy, and study its effect on accelerating neural functional recover.METHODS:The experiment was performed in the Department of Physiology of Shangqiu Medical College from September 2004 to February 2005.Sixtytwo 7day neonatal Wistar rats were divided randomly into three groups: ①rats in model plus acupuncture group(n=30) received acupuncture at Baihui,Quchi,Neiguan and Quanyong acupoints by using WQ10DI electronic acupuncture point therapeutic apparatus with 5 Hz frequency for 20 minutes and lasted for 7 days with 24hour interval 12 hours after hypoxiaischemia cerebral palsy;②rats in model group(n=20) were only modeled as hypoxiaischemia cerebral palsy models;③rats in control group(n=12) received no treatment.After 7day acupuncture,all rats were killed to detect the water content[(wet weightdry weight)/wet weight×100%] and cell apoptosis in brain tissue and the HPIAS1000 image analysis system was used to scan terminal deoxynucleotidyl transferase mediated dUTP nick end labeling sections to analyze the absorbency,and high absorbency indicated high expression). RESULTS:All 62 rats were involved in the result analysis.①The mean water in brain tissue was(79.3±2.5)%in control group,which was decreased in model plus acupuncture group as compared with that in model group[(82.2±1.9)%, (85.6±2.7)%,F=9.32,P < 0.05].②No apoptosis of neural cells in brain tissue was found in control group. The absorbency decreased in model plus acupuncture group as compared with that in model group(0.203±0.065, 0.254±0.071,t=2.916,P< 0.05). CONCLUSION:The mechanism of acupuncture in improving cerebral palsy may relate to inhibit the apoptosis of neural cells and decrease the cerebral edema.
