复方苦参注射液与5-氟尿嘧啶对大肠癌细胞凋亡的影响 被引量:23

摘要 目的探讨复方苦参注射液与5氟尿嘧啶对大肠癌LoVo细胞生长抑制及诱导凋亡的影响。方法台盼蓝染色法测定药物对LoVo细胞抑制作用与时间和剂量的关系,用荧光显微镜、琼脂糖凝胶电泳对细胞凋亡进行检测。结果台盼蓝实验表明,经40μg/L复方苦参注射液单独处理细胞,LoVo细胞增殖的抑制率24h,12.8%;48h,25.4%;72h,42.2%。12.5mg/L5Fu单独处理的LoVo细胞,其增殖抑制率24h,16.3%;48h,23.1%;72h,29.3%;而当两者合用时,对LoVo细胞增殖的抑制率24h,26.8%;48h,72.3%;72h,84.6%。复方苦参注射液和5Fu对LoVo细胞的生长均有抑制作用,并诱导发生凋亡。实验范围内其细胞凋亡成时间与剂量依赖关系。并且复方苦参注射与低浓度的化疗药物有协同作用,可产生较强的抑制细胞增殖的作用。结论复方苦参注射具有直接的抗肿瘤作用,并且具有增强5Fu化疗药物作用的功效。 Objective To investigate the effect of Matrine infections on LoVo cell and its synergetic effect with 5-Fu.Methods The proliferation inhibition of Matrine injections and its synergetic effect with 5-Fu on LoVo cells were detected by Trypanblue stain.Fluorescence microscopy and agarose gel electrophoresis were applied to study cell apoptosis of LoVo cells.Results Both Matrine injections and 5-Fu could inhibit the proliferation of LoVo cells seperately.The inhibited rate was 12.8% at 24 h, 25.4% at 48 h, 42.2% at 72 h respectively by 40 μg/L Matrine injection.The inhbited rate was 16.3% at 24 h, 23.1% at 48 h and 29.3% at 72 h respectively by 12.5 mg/L 5-Fu.While both of them used together,the inhibited rate was 26.8% at 24 h, 72.3% at 48 h and 84.6% at 72 h respectively.The results showed that Matrine injections had synergetic effect in inhibiting the proliferation of LoVo cells with low concentration of5-Fu.Conclusion Matrine injections has direct anticancer effect and can enhance the action of 5-Fu,and Matrine injections antagonize the toxicity of 5-Fu markly by decreasing the 5-Fu concentration.
