肥胖症患者皮下和网膜脂肪组织中PPARγ_2的表达与血浆瘦素、肿瘤坏死因子α和游离脂肪酸的关系 被引量:2

摘要 目的探讨过氧化物酶体增殖物激活受体γ2(PPARγ2)基因在中国汉族人腹部皮下脂肪和网膜脂肪组织中的表达水平,及与血浆瘦素(leptin)、游离脂肪酸(FFA)和肿瘤坏死因子α(TNFα)间的关系。方法选取28例非肥胖(BMI<25kg/m2)和19例肥胖症患者(BMI≥25kg/m2),采用RT-PCR方法检测大网膜与腹部皮下脂肪组织PPARγ2mRNA的表达水平,并测量身高、体重、腰围、臀围、收缩压、舒张压、空腹胰岛素、空腹血浆葡萄糖、血脂、瘦素、FFA和TNFα,计算胰岛素敏感指数和胰岛素抵抗指数(HOMA-IR)。结果(1)肥胖组血浆TG、VLDL-C、FINS、HOMA-IR、SBP、DBP、FFA、TNFα和瘦素均高于非肥胖组(P<0.05或P<0.01),ISI低于非肥胖组(P<0.01)。(2)肥胖组网膜和皮下脂肪组织的PPARγ2mRNA表达水平分别高于非肥胖组网膜下脂肪组织(P<0.01);肥胖组织内及非肥胖组织内的网膜与皮下脂肪组织PPARγ2mRNA表达水平差异无显著性(P>0.05)。(3)肥胖组、非肥胖及两组合并再分析显示,网膜和皮下脂肪组织PPARγ2mRNA表达水平与其他测量及计算指标均无明显相关性(P>0.05);瘦素、FFA、TNFα三者间及与其他指标也无明显相关性((P>0.05)。结论肥胖症患者网膜和皮下脂肪组织PPARγ2mRNA表达水平升高,并且血浆瘦素、FFA、TNFα的浓度较高。 Objective To investigate the peroxisomal proliferator-activated receptor gamma2(PPARγ_2)mRNA expression in abdominal subcutaneous and omental adipose tissues of obese subjects and their relation with plasma leption,FFA,TNF α level.Methods The PPARγ_2 mRNA expression level was measured by RT-PCR in abdominal subcutaneous and omental adipose tissues of 28 non-obese controls(BMI<25 kg/m^2)and 19 Subjexts(MBI≥25 kg/m^2.Anthropometric data such as height,body weight,waist circumference,hip circumference,plasma leption,FFA,TNFα were measured to calculate body mass index(BMI) and waist-hip ratio(WIIR).Systolic blood pressure(SBP) and diastolic blood pressure(DBP) were also measure.Fasting plasma insulin(FINS),glucose(FPG),lipids were determined.Insulin sensitivity index(ISI) and resistance index(HOMA-IR)were calculated.Results (1)Fasting plasma TG,VLDL-C,SBP,DBP,insulin,leption,FFA,TNF α and HOMA-IR were significantly higher in obese subjects than those in non-obese controls ((P<0.05 or P<0.01).Insulin sensitivity index was significantly decreased in obese subjects than that in non-obese controls(P<0.01).(2)The PPARγ_2 mRNA expression in obdominal subcutaneous and omental adipose tissue were significantly higher in subjects with obesity than those in non-obese controls(P<0.01).There were on differences in the expression of PPAR γ_2 mRNA between omental and abdominal subcutaneous adipose tissue among intra-obese group and non-obese control(P>0.05).(3)There was not significantly correlation between expressions of PPAR γ_2 mRNA in abdominal subcutaneous no omental adipose tissue and other parameters(P>0.05).There were not significantly correlation between leptin,FFa,TNF α and other parameters(P>0.05).Conclusion The PPARγ_2 mRNA expression in abdominal subcutaneous and omental adipose tissue are significantly higher in subjects with obesity.Fasting plasma leptin,FFA,TNF α are significantly higher in obese subjects than that in non-obese controls.There are not significantly correlation are expressions of PPARγ_2 mRNA in abdominal subcutaneous and omental adipose tissue with leptin,FFA,TNF α.
