摘要 目的:为了给脂溶性的抗肿瘤药物筛选高溶解性、低细胞毒性的有机溶剂。方法:采用MTT法,研究了丙酮、甲醇、乙醇、DMSO(二甲基亚砜)、异丙醇及甘油等6种有机溶剂对U937(人白血病细胞株)、95D(猪肺癌细胞株)及AGS(人胃癌细胞株)三种细胞株的细胞毒性。结果:丙酮对三种细胞株的毒性较弱,在10%的体积分数时也没有明显的毒性,是一种良好的有机溶剂,甲醇在使用时体积分数应低于1%;对U937细胞,乙醇、甘油、异丙醇、二甲基亚砜(DMSO)的IC50分别为6.28%、5.98%、6.41%、14.18%,甲醇对U937细胞不成线性关系,在体积分数<1%时没有明显的细胞毒作用,当浓度达到3%以上时表现出明显的细胞毒作用;对95D细胞,甲醇、乙醇、甘油、异丙醇、DMSO的IC50分别为9.36%、3.23%、7.22%、1.13%、2.15%,对人AGS细胞株,甲醇、乙醇、甘油、异丙醇、DMSO的IC50分别为9.27%、2.52%、7.22%、1.13%、2.15%;结论:丙酮是脂溶性抗肿瘤药物的首选有机溶剂。 Objective: To select organic solvents for dissolve some compounds which have bad water-solubility in screening antitumor agents. Methods: The cytotoxic effect of six organic solvents, such as acetone, methanol, ethanol, glycerin,isopropylalcohol and DMSO,on three tumor cell lines (95D,AGS and U937)were studied by MTT. Results: 1 Acetone is the best in the six organic solvents, it has little cytotoxic effect even its volume proportion is 10% to each three tumor cell lines. The volume proportion of Methanol should less than 1% if Methanol will be used;2 The IC_(50)of ethanol, glycerin,isopropylalcohol and DMSO on U937 are 6.28%,5.98%,6.41%,14.18%,Acetone has little cytotoxic effect when its volume proportion less than 1%,but marked when its volume proportion more than 3%. The IC_(50)of acetone, ethanol, glycerin,isopropylalcohol and DMSO on 95D are 9.36%,3.23%,7.22%,1.13%, 2.15%. The IC_(50)of acetone, ethanol, glycerin,isopropylalcohol and DMSO on AGS are 9.27%, 2.52%,7.22%,1.13%,2.15%. Conclusion: Acetone is the best organic solvent to the bad water-solubility antitumor agents.
