摘要
采用改良的CTAB法提取葡萄幼叶,得到了可用于RAPD分析的DNA。通过对反应体系和反应程序的筛选,建立了适合葡萄基因组RAPD分析的技术体系。并运用该体系对供试的29个鲜食葡萄品种进行了聚类分析。
The improved CTAB method was used to extract Vitis genomic DNA, and well-purified DNA was obtained and qualified for RAPD-PCR in this study.Different PCR programs and the ingredients in RAPD reaction system were experimentally selected and an optimal program of the RAPD analysis for vitis genomics was settled. The twenty-nine species could be identified by the amplified bands from the 3 primers employed in this experiment.
