摘要
目的:探讨运动性低血睾酮与血清皮质醇(酮)水平之间的关系,以及益气补肾中药对二者是否具有干预作用。方法:50只Wistar大鼠随机分为安静对照组(n=10)、安静服药组(n=10)、训练组(n=15)和训练服药组(n=15)。经6周递增负荷游泳训练后,使用放免法和酶免法分别测定大鼠血清睾酮和皮质酮水平;利用RT-PCR方法检测大鼠睾丸11β-HSDmRNA的表达水平。结果:(1)训练组大鼠血清睾酮水平显著降低,而训练服药组大鼠的血清睾酮水平则未降低;(2)各组之间的血清皮质酮浓度和11β-HSDmRNA的表达水平没有显著性差异。结论:(1)研究结果提示,长期耐力训练后造成大鼠的血清睾酮水平下降,而训练后糖皮质激素-皮质酮没有升高现象,同时睾丸11β-HSDmRNA的表达亦没有变化,提示运动性低血睾酮可能与皮质酮和11β-HSD之间的相互调节无关;(2)益气补肾中药对大鼠长时间运动后的低血睾酮有干预作用,但对血清睾酮皮质酮和11β-HSDmRNA的表达均没有效应,说明该中药在此方面没有调节作用。
Objective:The relation between exercise-associated low-level serum testosterone and corticosterone concentration as well as Chinese Medicine Compound for Invigorating Qi and Tonifying the Kidney on these in rats were observed.Methods:50 Wistar Rats were randomly divided into 4 groups: sedentary control group (SC, n=10), sedentary with medicine group (SM, n=10), training group (T, n=15) and training with medicine group(TM, n=15). After 3 and 6-week swimming, serum testosterone and corticosterone level of rats were tested through radioimmunological and EIA methods, respectively; Using RT-PCR, we measured testicular 11β-HSD mRNA levels of rats. Results: (1)In T group, serum testosterone level was lower than that in SM and TM groups. On the contrary, serum testosterone concentration of TM group was not decreased than that of SC group. (2)Whether the Chinese medicine was fed or not, serum corticosterone concentration and the expression of 11β-HSD mRNA of training groups were not markedly increased than that of sedentary groups.Conclusion:(1)The consequence of serum corticosterone concentration and 11β-HSD mRNA levels suggest that they may be not involved in exercise-associated low-level serum testosterone.(2)Through improving serum testosterone level,Chinese medicine compound of 'invigorating 'qi'and tonifying the kidney' could be effect on preventing and curing the generation of low-level serum testosterone during strenuous training. But this Chinese medicine compound could not effect on serum corticosterone and testicular 11β-HSD mRNA level.
