摘要
目的:探讨双黄连及其含药血清对呼吸道合胞病毒(RSV)感染的气道上皮细胞(A549)的炎症因子释放的影响及其药物作用机理。方法:以ELISA法测定炎症介质的含量,观察复方双黄连及其含药血清以不同的给药方式(Ⅰ组:感染前给药、Ⅱ组:感染时给药、Ⅲ组:感染后给药)对RSV刺激A549细胞引发的炎症介质IL8、IL6、TNFα释放的作用。结果:RSV刺激气道上皮细胞A549后IL8的分泌水平增加,而对IL6、TNFα影响不大;双黄连复方及其含药血清均能显著降低RSV诱导A549释放的IL8水平,并以感染前给药的途径下调IL8的作用最为明显。结论:RSV能诱导气道上皮细胞释放炎症因子IL8,双黄连可通过抑制RSV引起的IL8释放,并在病毒感染的气道炎症控制中起着一定的作用。
Objective:To study the effect of Shuanghuanglian (SHL) and SHL-containing serum on the release of chemokine in A549 airway epithelial cells activated by respiratory synytial virus (RSV) and its anti-virus mechanism. Methods:Using cell-culture technique to cultivate A549 cells, designing three drug addition groups (group Ⅰ: adding pre-infection; group Ⅱ: infection; group Ⅲ: post-infection) to investigate the release of chemokine in RSV-infected A549, collecting supernatants and using ELISA to detect the concentration of chemokine, IL-8, IL-6, TNF-α.Results:RSV can activate airway epithelial cells to release more IL-8, but show no effect on IL-6 and TNF-α. SHL and SHL-containing serum can decrease the level of IL-8 in RSV-infected A549, especially addiction I.Conclusion: RSV can stimulate the secretion of chemokine in A549 cells, but SHL and SHL-containing serum can reverse RSV-induced increases in IL-8 and take a role in controlling the airway inflammation of RSV infection.
