摘要
目的 通过研究大鼠急性脊髓损伤后基质金属蛋白酶 9(MMP 9)表达的时空变化,探讨基质金属蛋白酶9在急性脊髓损伤后的作用。方法 使用改良Allen法制作大鼠急性脊髓损伤模型,检测脊髓损伤后基质金属蛋白酶 9在损伤脊髓中表达的时空分布特点。结果 损伤组急性脊髓损伤后1小时,可见基质金属蛋白酶 9的表达,损伤后1~2天达到高峰,7天仍有表达。阳性细胞主要表达在缺血神经元、血管内皮细胞和嗜中性粒细胞。对照组未见基质金属蛋白酶9表达。结论 基质金属蛋白酶 9在脊髓损伤后的早期表达,提示基质金属蛋白酶9可能参与脊髓损伤后水肿形成和炎性细胞的浸润,加重了继发性脊髓损伤。
Objective To investigate the early expression of matrix metalloproteinase-9 after actue spinal cord injury.Methods The models of spinal cord injury in rats were induced with Allen's way and the expression of matrix metalloproteinase-9 in injuried spinal cord was observed.Results The expression of matrix metalloproteinase-9 increased 6h after spinal cord injury,which reached its peak 1,2 days after spinal cord injury(SCI).The matrix metalloproteinase-9 positive cells were mainly expressed on ischemia neuron, angioendothelial cells and neutrophile.The express of matrix metalloproteinase-9 wasn't found in the sham-operated rats.Conclusion The early matrix metalloproteinase-9 may be partially involved in the secondary pathogenesis of spinal cord injury.
