细胞凋亡及其相关因子在HAM/TSP中的表达及其意义

摘要 目的研究人嗜T淋巴细胞病毒Ⅰ型相关性脊髓病/热带痉挛性瘫痪(HAM/TSP)外周血淋巴细胞凋亡情况及凋亡相关因子Fas、肿瘤坏死因子α(TNFα)和Bcl2蛋白在HAM/TSP发生发展中的作用。方法应用单细胞电泳(彗星电泳)检测8例HAM/TSP患者(HAM/TSP组)、8例其他神经系统感染性疾病(OIND)患者(OIND组)和8名健康献血者(健康对照组)的外周血淋巴细胞的凋亡情况,并应用流式细胞术对淋巴细胞上Fas、TNFα和Bcl2蛋白进行定量检测。结果HAM/TSP组外周血淋巴细胞凋亡率明显增高,与健康对照组比较差异有极显著性(P<0.01);淋巴细胞上Fas、TNFα表达水平与OIND组及健康对照组相比显著增高(均P<0.01),Bcl2表达较OIND组显著降低(P<0.05)。结论淋巴细胞凋亡及凋亡相关因子Fas、TNFα和Bcl2蛋白可能在HAM/TSP的发生发展中起重要作用。 Objective To study the apoptosis of peripheral blood lymphocyte cells in patients with HAM/TSP and the roles of Fas, TNF-α and Bcl-2 in occurrence and progression of HAM/TSP.Methods The single cell gel electrophoresis (comet assay) was used to detect the apoptosis of lymphocyte cells in 8 patients with HAM/TSP, 8 cases with other inflammatory neurologic diseases (OIND) and 8 healthy blood donors. The Fas, TNF-α and Bcl-2 protein expressions were quantitatively determined by flow cytometry from 8 cases of HAM/TSP.Results The apoptotic ratio of lymphocyte cells in patients with HAM/TSP significantly increased compared with the controls (P<0.01). Patients with HAM/TSP had higher Fas and TNF-α levels than those with OIND and controls (all P<0.01). Significant decrease of Bcl-2 from patients with HAM/TSP was shown compared with OIND (P<0.05). Conclusion Apoptosis of lymphocyte cells and apoptotic correlation factors including Fas, TNF-α and Bcl-2 may play an important role in the development of HAM/TSP.
