摘要
以试管苗的小叶为外植体,探讨了月季品种‘萨蔓莎’愈伤组织诱导及植株再生的方法。结果表明:高浓度的生长素能诱导小叶产生愈伤组织,经24D7.0~10.0mg/L光下诱导愈伤15d的小叶外植体,在含TDZ1.5mg/L的MS培养基上光下培养约20d后有白色假珠芽形成,此芽暗培养在含NAA、ZT和GA3的培养基上产生新的假珠芽和新的愈伤组织,通过持续选择继代生活力高的组织,约1年后获得可长期继代的愈伤组织,到目前为止已保存了16个月。此愈伤组织在TDZ1.0mg/L、NAA0.01mg/L和GA30.1mg/L的诱导下30d内分化出不定芽,在含BA2.0mg/L和NAA0.01mg/L的培养基上不定芽伸长形成正常植株。
White pseudobulbils were initiated from in vitro grown leaf explants of Rosa hybrida‘Samantha’following an induction period of 15 d on MS basal medium supplemented with 24-D 7.0-10.0 mg/L and a subculture of 20 d on MS medium with TDZ 1.5 mg/L under light. The pseudobulbils were subcultured on medium with NAA, ZT and GA_3,and during the process, new pseudobulbils and new callus were formed. After a year of continuous selection for fresh and nodular callus,callus that could be continuously subcultured was obtained, and has been maintained for 16 months till now. Adventitious buds could be induced from the callus on MS medium with TDZ 1.0 mg/L, NAA 0.01 mg/L and GA_3 0.1 mg/L and elongated into shoots on MS medium with BA 2.0 mg/L and NAA 0.01 mg/L.
