力生长因子

摘要 力学刺激能调控肌肉、骨骼等组织的基因表达,从而引起这些组织结构和功能的变化,如肌肉肥大、骨骼强壮等。在这些组织中,胰岛素样生长因子-1(IGF-1)是力学敏感因子。对骨骼肌进行的力学拉伸实验发现,力学刺激导致肌肉肥大的同时伴有2种IGF-1变构体的产生。这2种变构体是由于IGF-1基因插入突变和选择性剪切产生,分别命名为力生长因子(mechanogrowthfactor,MGF),肌肉型IGF-1(IGF-1Ea)。MGF能激活卫星细胞而使成肌细胞增殖,IGF-1Ea则促使成肌细胞分化形成肌管。进一步研究发现,在萎缩的肌肉中,这2种因子的产生发生障碍。
