高效液相色谱法测定螺旋藻中几种色素的研究 被引量:3

摘要 目的建立螺旋藻中主要色素成分(叶绿素-a、叶绿素-b、叶黄素、β-胡萝卜素和新叶黄素)的高效液相色谱快速分析方法.方法螺旋藻中的色素用90%的丙酮提取,以安捷仑ZORBAX Stable Bound(2.1×50 mm,1.8μm)色谱柱为固定相,四氢呋喃和水梯度洗脱为流动相,用二极管矩阵检测器检测.结果5种色素在2.0min内分离,标准回收率为98%~102%,相对标准偏差为1.1%~1.4%.结论本方法样品前处理简便、快速,分析时间比常规色谱柱大大缩短,较常规色谱柱流动相消耗减少,结果满意. ObjectiveTo establish a high performance liquid chromatography (HPLC) method for the determination of pigments (chlorophyll-a, chlorophyll-b, β-carotene, lutein,neoxanthin)in springer.MethodThe pigments were extracted from the sample with 90% of acetone.Then,the pigments was separated on a ZORBAX Stable Bound (2.150 mm, 1.8 μm) column, with THF and water as mobile phase at a flow-rate of 0.3 ml/min, and monitored with the photodiode array detector at 450nm.ResultThe four pigments were separated completely within 2.0 min.The recoveries of the pigments were 98%102%,and relative standard deviations were 1.1%1.4%. Results are satisfactory.ConclusionIn this method,the sample preparation is simple.The separation time greatly shorten and solvent consumption is saved.The results are satisfactory.
