中药西红花指纹图谱研究

Studies on the Fingerprint of Saffron

目的应用高效液相色谱法对上海、浙江、江苏三大产地的药材进行全面质量考察。方法采用ShimadzuVP-ODS柱(150mm×4.6mm,5μm),流动相甲醇-水(55∶45),流速1ml·min-1,柱温30℃,检测波长440nm。结果10批西红花药材得到的色谱指纹图谱有8个共有峰,相似度均大于0.95。结论本文可提供清晰的HPLC指纹图谱,图谱特征性及专属性强,可结合含量测定用于全面控制药材质量,确保每批产品的稳定性。实验方法简便、准确、重复性好。

OBJECTIVE High performance liquid chromatograph (HPLC)was applied for reviewing quantity of saffron,which comes from Shanghai、Zhejiang and Jiangsu province.METHODS HPLC analysis was performed on a Shimadzu VP-ODS column(150mm×4.6mm,5μm),with the mixture of methanol-H_2O(55∶45)as mobile phase.The temperature was set up at 30℃ and the flow-rate was 1ml·min -1 .The wavelength of detection was 440nm. RESULTS The HPLC chromatographic fingerprint of saffron,showing 8 characteristic peaks,was established from 10 lots of herb of saffron.The similarity of HPLC fingerprint is above 0.95.CONCLUSION The clear HPLC fingerprint was obtained.The chromatographic fingerprint of total glucosides from saffron with high specificity can be used to control its quality and assure lot-to-lot consistency.The standard fingerprint of the saffron tas been gotten.The method is simple and reliable.
