摘要
目的:观察类风湿性关节炎患者单核/巨噬细胞用GM-CSF刺激活化前后,基质金属蛋白酶(MMP-1,MMP-3)合成及CD147表达的变化。方法:从10例类风湿性关节炎患者患者外周血及滑液分离单核细胞,同时取10例健康献血员外周血单核细胞为对照。分离的单核细胞在RPMI1640培养液中培养,用GM-CSF刺激,用ELISA法测定刺激24h,48h,72h后MMP-1,MMP-3的含量。用流式细胞术测定刺激前后细胞表面CD147表达量。结果:GM-CSF刺激后,细胞呈多形性的巨噬细胞样改变,MMP-1,MMP-3合成增加,而且这种刺激作用与GM-CSF的剂量相关,100ng/ml时刺激作用显著高于12.5ng/ml及25ng/ml,而与作用时间无显著相关性;滑液单核细胞MMP-1,MMP-3表达水平高于外周血单核细胞。GM-CSF刺激后,单核细胞表面CD147的表达显著高于刺激前;滑液单核细胞表达量高于外周血。结论:,RA患者外周血及滑液单核细胞用GM-CSF刺激活化后MMP-1,MMP-3合成增加,CD147表达亦增加,MMPs合成增加可能与CD147表达增加相关。
Objective: To study the variation of matrix metalloproteinases-1,3 production and CD147 expression by monocyte/macrophage from peripheral blood and synival fluid of rheumatoid arthritis patients after stimulating with GM-CSF. Methods: Monocyte was isolated from peripheral blood and synival fluid of rheumatoid arthritis patients with Ficoll-Hypaque, then incubated in RPMI1640 with different amounts of GM-CSF. The contents of MMP-1,MMP-3 were evaluated by enzyme-linked immunosorbent assay (ELISA) in 24h,48h.and 72h respectively. CD147 expressed on the surface of monocyte/macrophage was assessed by flow cytometry. Conclusion: The production of MMP-1,MMP-3 and CD147 expression by monocyte from peripheral blood and synival fluids of RA patients were enhanced significantly when stimulated with GM-CSF, and the stimulating effects of GM-CSF on MMP-1,MMP-3 production by monocyte maybe correlated with the increased expression of CD147 on the surface.
