假性湿疣组织HPV6bDNA相关序列的检测及其电镜观察

摘要 本文用光敏生物素标记人乳头瘤病毒6b型(HPV6b)DNA探针进行斑点杂交,检测12例假性湿疣组织中HPV6bDNA相关序列。结果6例杂交阳性,10例正常宫颈组织杂交阴性。对9例假性湿疣做了电镜观察,结果组织有增殖型和蜂窝样空泡变型两种改变。9例中有4例与HPV6bDNA探针杂交阳性,其中1例为增殖型,8例为蜂窝空泡变型,并有1例观察到病毒颗粒与晶格。 The relationship between human papillomaviruses(HPV) infection and pseudocondyloma occurrence was studied in this paper. The DNA were isolated from the pseudocondyloma tissue of 12 cases who had been pathologically diagnosed as the patients with pseudocondyloma. Then the dotted hybridization was carried out with photobiotinated HPV 6b DNA probes to detect the HPV homologous sequences in the DNA samples.The results showed 6 of 12 samples were hybridized with the probe. The hybridization positive rate was 50%. But the other 10 DNA samples(control) from normal cervical tissue were never hybridized with the HPV 6b probe. Further more,the 4 samples from the tissues hybridized with HPV 6b were observed under SEM and TEM,The results suggested that there were 2 different types of morphology,One was the proliferative type with 1 case which just liked the 5 cases of the hybridization negative samples. And anotherone was vacuolar type with 3 cases. The virus particles and crystal were observed in the nuclei of one of the 3 vacuolar cases. In conclusion,the results of the paper supported the hypothesis that the occurrence of pseudocondyloma was relative to the infection of HPV.
