摘要
目的:检查Ang-1/-2蛋白在小鼠胚胎着床期子宫内膜的分布及mRNA的表达,以研究Ang-1/-2基因在胚胎着床过程中所起的作用和生物学意义。方法:取D2,D4,D6和D8昆明种小鼠子宫内(蜕)膜,分别用免疫组织化学S-P法和原位杂交技术,检查着床期子宫内膜中Ang-1/-2蛋白的表达及其mRNA的转录水平。并用计算机图像分析技术检测不同时期子宫内膜中Ang-1/-2表达的平均光密度值。结果:免疫组化显示,Ang-1蛋白在基质细胞、上皮细胞和血管壁中表达,而Ang-2蛋白则在腺上皮细胞及基质细胞中表达,两者随着妊娠天数的增加而表达增强(P<0·01)。原位杂交显示,Ang-1mRNA自D2起即表达于基质细胞中,D4血管壁胞浆中也出现阳性表达;而Ang-2mRNA自D2起则表达于基质细胞和腺上皮,D4血管壁胞浆中也出现阳性表达。Ang-1/-2mR-NA表达强度在D6,D8逐渐增加。平均光密度值差异有显著性(P<0·01)。结论:Ang-1/-2基因在小鼠胚胎着床期血管新生和血管重塑过程中起重要作用,从而有助于囊胚成功着床。
Objective:To investigate the expression and distribution of angiopoietin-1/-2(Ang-1/-2) in the endometrium of early pregnant mice. Methods:The endometrium of 2-,4-,6-,and 8day pregnant Kunming mice were observed.The expression of protein and the transcription level of mRNA of Ang-1/-2 were detected by immunohistochemical staining and in situ hybridization methods respectively.Image analysis system was used to measure the average optical density of Ang-1/-2 in endometrium of different stages of pregnancy and the controls.Results:By immunohistochemical staning,it was showed that Ang-1 was expressed in the stromal cells,the luminal epithelium and vascular endothelium.Ang-2 localized in the cytoplasma of glandular epithelium and stromal cells.The expression of Ang1 and Ang2 was gradually increased through the process of pregnancy (P<0.01).In situ hybridization showed that Ang-1mRNA was detected in stromal cells at d2,hybridization signal localized in both stromal cell and vessel epithelial cell of d4.Ang-2 mRNA was observed in stromal cells and glandular epithelium at d2,and also positive in cytoplasm of vascular endothelial cells of d4.The intensity of Ang-1/-2 mRNA expression was increased from d6 to d8 of pregnancy.The average optical density had significant difference (P<0.01). Conclusion:Hyperexpression of Ang-1/-2 in pregnant mouse endometrium indicates that Ang-1/-2 may play an important role in the process of vascularization and vascular remodeling during embryo implantation,which is helpful for the successful implantation.
