纹状体提取液增强体外培养的中脑黑质神经元的发育 被引量:10

摘要 制备新生2d龄大鼠纹状体提取液及其不同分子量的组分,用快速自动比色微量分析法检测它们对体外培养的14d龄大鼠胚胎中脑黑质神经元的影响。结果表明,纹状体提取液有支持中脑黑质神经元生存及增强其活性的作用,而且具有剂量依赖关系,其最适浓度为1.25μg/ml。纹状体提取液还能促进中脑黑质神经元对2H-DA和3H-GABA的摄取,同对照组相比,有显著性差异(P<0.01)。通过检测纹状体提取液小于10kD、10~30kD和大于30kD3种不同分子量组分对中脑黑质神经元的作用,证明有活性的部分是分子量为10~30kD的组分。 The diffusible trophic factors released by neural target tissues can enhance survival and development of neurons.The striatum is a major target tissue of mesencephalic substantia nigra(SN)neurons.We have prepared extracts of striatum from 2-day-old rat brain and various molecular weight fractions of striatal extracts,Effects of these extracts on the development of cultured mesencephalic SN neurons from embryonic day l4 rat brain were measured by a rapid automated colorimetric microassay(MTT colorimetric microassay).The experimental results show that striatal extracts can enhance the survival and activity of mesencephalic SN neurons.These effects are dose-dependent, with optimal stimulation occurring at a concentration of l.25μg/ml.The ability of mesencephalic SN neurons to take up 3H-DA and3 H-GABA is increased by striatal extracts(compared to control,P<0.0l).To measure the effect of various molecular weight fractions(<10kD,10~30kD,and>30 kD)of striatal extracts on the development of mesencephalic SN neurons,having obtained the active fraction is that of l0~30 kD.
