摘要
用光镜免疫组织化学和原位包埋免疫电镜技术观察了低氧对成年大鼠肺降钙素(calci~tonin,CT)阳性神经内分泌细胞的影响。计数了单位面积内单个神经内分泌细胞(NEC)、神经上皮小体(NEB)以及NEC与NEB细胞个数之和的阳性细胞总数(TNE)。统计分析结果显示,低氧导致TNE、NEB和组成NEB的细胞数目增多。原位包埋免疫电镜法发现,降钙素免疫反应物定位于神经分泌颗粒内。
By means of immunohistochemical PAP method for calcitonin in both light microscopy and electron microscopy,the influence of hypoxia on solitory neuroendocrine cells(PNEC )of adult rats was studied.The number of solitary neuroendocrine cells(NEC)and neuroepithelial bodies(NEB )per unit area(cm2)were calculated separately.The results showed : The positive immunoreactivity for calcitonin was demonstrated in the N EC and NEB in both normoxic and hypoxic rats,There were much more calcitonin positive cells among the TNE and NEB in hypoxic rats and showed significant difference statistically(P<0. 05 ). Furthermore,we proved that the mean number of the calcitonin positive cells per NEB was also more in the hypoxia group than in normal(P<0.02).However,the NEC did not show apparent difference between the two groups.Such findings seem to suggest that the increase in the calcitonin positive TNE in the hypoxic group ascribed to the increase of calcitonin positive cells per NEB and indicate that pulmonary neuroendocrine cell is heterogeneous in response to hypoxia. In addition,we also confirmed that the calcitonin immunoreactivity is localized in neurosecretory granules by immunoelectron microscopy.
