摘要
用免疫组织化学和免疫电镜双标记法研究了胰岛淀粉样多肽(IAPP)免疫反应细胞在正常大鼠胃肠胰系统的分布、定位和形态特征,并观察了IAPP与胰岛素在胰腺的共存。结果表明,大鼠胰腺和胃肠道各段均见有IAPP免疫反应细胞。在胰腺,IAPP与胰岛素共存于胰岛的B细胞内,也共存在散在于外分泌部的胰岛素免疫反应细胞内。胃肠道粘膜内IAPP免疫反应细胞主要存在于上皮和固有膜。它们的数量以胃和十二指肠较多,结肠较少。本研究首次发现IAPP免疫反应细胞在空肠、回肠和结肠内均有分布。这为进一步探讨IAPP在调节糖代谢和协调胰岛素分泌及通过刺激胃议素调节胰腺外分泌功能提供了形态学依据。
In this study, light microscopic immunohistochemical and immunoelectron microscopic double-labeling techniques were used to observe the distribution,localization and morphology of IAPP immunoreactive cells in adult rat gastroenteropancreatic system,and to explore the coexistence of IAPP with insulin in pancreas,The results showed that IAPP immunoreactive cells were found in pancreas,stomach,duodenum,jejunum,ileum,colon and rectum of rat.In pancreas IAPP coexistence with insulin both in the secretory granules of islet B cells and in insulin immunoreactive cells dispersed in exocrine pancreas was observed;In gastrointestinal tract IAPP immunoreactive cells were located in epithelium and lamina propria,The number of IAPP immunoreactive cells was more in stomach and duodenum but less in colon,The distribution of IAPP immunoreactive cells in jejunum,ileum and colon was reported for the first time. The results provided morphologic evidences for further exploration of the physiological effects of IAPP on regulation of glucose metabolism and coordination of insulin secretion.
