体外LAK细胞杀伤人直肠腺癌细胞时的细胞骨架变化 被引量:1

摘要 应用免疫荧光细胞化学和透射电镜观察体外LAK细胞杀伤人直肠腺癌细胞(HR_(8348))时效靶细胞内细胞骨架的变化。结果表明:两种细胞相互识别后均出现趋向运动,趋向运动的细胞突起内含有丰富的微丝,微丝的排列与细胞运动的方向一致。效靶细胞相互接触后,两种细胞浆内的微管均出现重排现象。两种细胞质膜接触处可见成束的微丝及微管,微丝及微管集中处含有丰富的溶酶体颗粒。微丝及微管抑制试验均不出现上述变化。结果提示:微丝可能在效靶细胞趋向运动及连接中起作用,效靶细胞接触后微管的重排可能与靶细胞的溶解有关。 Using immunofluorescence cytochemistry and transmission electron microscopy, changes of cytoskeleton of the interaction between LAK cells and HR8348 cell were observed in vitro. The results showed that effector and target cells presented chemotatic moving after recognizl I each other. There were abundant miercfilament (MF) where the arrangement of MT was tonsistent with the longitude of cell pro-ness in the projections of chemotactic moving cell. The rearrangement of microtubuies (MT) within effector-target cells occured following cell contact. There are abundant MF and MT in the side of effector-target cell binding site. MT accumulated sites contained more lysosmes. With adding MF and MT inhibitor, There are no the attachment of MF and MT rearrangement between effector-target cells. The results suggested that MF may play a role in chemotactic moving and effector-target cell binding, while the rearrangement of MT may lead to lysis of targets.
