环氧合酶-2在早孕期对胚胎滋养层细胞凋亡的抑制作用的研究

摘要 目的:探讨环氧合酶-2(COX-2)在早孕期对胚胎滋养层细胞凋亡的抑制作用及相关途径。方法:用免疫组织化学的方法观察早期正常胚胎滋养层细胞和难免流产胚胎滋养层细胞各20例中COX-2和Bcl-2蛋白表达的差异;建立人妊娠早期滋养层细胞体外培养体系,制作细胞爬片,并用COX-2的选择性抑制剂Nimesulide以100μmol/L浓度处理24h,免疫细胞化学的方法比较对照组和处理组COX-2和Bcl-2蛋白表达的差异。结果:难免流产患者胚胎滋养层细胞较正常者COX-2和Bcl-2蛋白表达均明显降低;用COX-2的选择性抑制剂Nimesulide处理的滋养层细胞较对照组COX-2和Bcl-2蛋白表达也明显降低。结论:COX-2在早孕期可抑制胚胎滋养层细胞的凋亡,部分是通过Bcl-2家族途径。 Objective:To investigate the potential role of cyclooxygenase-2 in inhibiting apoptosis of human trophoblasts during early pregnancy and its possible mechanism.Methods:Immunohistochemical staining was used to detect the different expression of COX-2 and Bcl-2 in trophoblasts between normals and the inevitable abortion.Human trophoblasts of early gestation were cultured in vitro.The cells were treated with COX-2 specific inhibitor nimesulide with the concentration of 100 μmol/L for 24 hours,then immunocytochemical assessment was performed for alterations in expression of COX-2 and Bcl-2.Results:COX-2 and Bcl-2 expression were obviously(P<0.01) higher in the normals than those inevitable abortion.A significant decrease in COX-2 and Bcl-2 compared with controls in the cell lines(P<0.01) after treatment with the COX-2 specific inhibitor was observed.Conclusion:COX-2 could inhibit apoptosis of human trophoblasts during early pregnancy,partly through the Bcl-2 family pathway.
