反相高效液相色谱法测定血浆中青霉素浓度 被引量:4

摘要 目的:建立测定血浆中青霉素浓度的高效液相色谱法。方法:色谱柱为C18,以0.05mol/L磷酸盐缓冲液-乙腈(75∶25)为流动相,青霉素V为内标。血浆样品经C18SPE小柱纯化、衍生化后进样,在325nm波长处检测,按内标法定量。结果:青霉素检测浓度线性范围为0.05~20μg/L,最低检出浓度为0.05μg/L,日内RSD<2.5%,方法回收率在95%~104%范围内。结论:本法具有快速、简便、灵敏、准确等优点,可用于青霉素血药浓度测定。 OBJECTIVE:To establish the method to determine Penicillin G in human plasma by RP-HPLC.METHODS:C 18 was used as the chromatographic column.The mobile phase consisted of 0.05 mol/L phosphate buffer solution-acetonitrile(75∶25)with Penicillin V as the internal standard.The plasma sample introduction was carried out after purified and derivatized by C 18 SPE.The determination was carried out when the wavelength was at 325nm.The quantitation was done by internal standard method.RESULTS:Linear concentration range for penicillin was 0.05~20μg/L;the concentration limit was 0.05μg/L;the intraday RSD was less than 2.5% and the method recovery rate was within the range of 95%~104%.CONˉCLUSION:This method was simple,rapid,sensitive and accurate,which can be used to determine the blood concentration of penicillin.
