摘要
目的:了解慢性髓细胞性白血病患者肿瘤细胞VEGF、b-FGF、KDR、Flt-1基因表达,探讨血管生成因子与肿瘤细胞间的相互作用。方法:用RT-PCR方法检测16例慢性髓细胞性白血病患者骨髓粒系细胞VEGF、b-FGF、KDR、Flt-1基因表达,以5例原发性血小板减少性紫癜患者作对照组进行比较。结果:VEGF和b-FGF基因高表达,与对照组比较有显著差异(P<0.05)。KDR基因基本不表达,Flt-1明显表达但与对照组无明显差异。结论:虽然KDR和Flt-1的表达意义不大,但VEGF和b-FGF基因高表达。证实了肿瘤细胞分泌血管生成因子增加,可能存在自分泌途径。
Objective: We studied the gene expression of VEGF, b-FGF, KDR and Flt-1 in 16 CML pateins in order to acknowledge the reciprocity between Angiogenesis factors and Leukaemia cells. Methods: Myeloid cells were isolated from 16 CML patients as study group and 5 ITP patientsas control group to detect the gene expression of VEGF > b-FGF, KDR and Flt-1. Results: The gene expression of VEGF and b-FGF were both higher than those of control group(P<0. 05). KDR gene was hardly expressed in study group, while Flt-1 were highly expressed in both groups,and the difference has no statistics significance. Conclusion: Although the expression of KDR and Flt-1 genes had no obvious correlation according to our study,the high expression of VEGF and b-FGF genes show that the high secretion of the Angiogenesis factors in leukaemia cells may be caused by outocrine mechanism.
