丹参注射液对鸡胚额骨分离细胞培养生长影响的组织化学观察成骨细胞样细胞的发育与成熟

Effect of Radix Salviae Miltiorhizae on Growth of Isolated Cells from Embryonic ChickenFrontal Bone Cultured in Vitro I.The Develop-ment and Maturation of Osteoblast -like Cells

实验用鸡胚额骨分离细胞体外培养结合组织化学技术观察丹参注射液对成骨细胞样细胞增殖、分化和成骨功能的影响。发现：（１）丹参能明显促进成骨细胞样细胞的分裂和增殖。（２）丹参能促进成骨细胞样细胞提前出现伪足伸缩运动和胞外吐作用，使丹参组的培养液内出现大量细小漂浮性颗粒，该颗粒对酸性粘多糖染色呈阳性反应。（３）成骨细胞样细胞经发育后能分泌直径约为３ｕｍ的小光状颗粒。这种颗粒显酸性粘多糖着色（早期），胶原和钙盐着色（晚期）在细胞集落周围聚集、融合。形成骨小结节和骨小片，并愈积愈大。经四环素活体标记后在荧光显微镜下呈金黄色，因此是新生骨组织。丹参组织形成的骨小结节和骨小片在数量上和体积上明显大于对照组。

AbstractIsolated osteoblast-like cells from embryonicchicken frontal bone were cultured in vitro and his-tochemical methods were usedto o6serve thedfectof Radix Salviae Miltiorrhizae (R.S.M.) on theirproliferation,differentiation and osteogenic capaci-ty. The results revealed that:(1)the mitosis andproliferation of the osteoblast-like cells could beaccelerated by R.S.M.;(2)the pseudopodiastretched out and drew back actively and exocyto-sis in osteoblast-lide cells could be accelerated byR.S.M., yielding a large number of floating parti-cles in the extracellular medium in the group givenR.S.M. These particies revealed positive acid mu-copolysaccharide staining;(3) after development,the osteoN8st-like ecells could secrete 3micron-diameter vesicular particles,whichcouldshow positive acid mucopolysaccharide staining inthe early stage,and then positive collagen and cal-cium salt staining in the later stage. they could ag-gregate and fuse around the cell colonies to formbone nodules andnakes.when they were labeledwith Tetracycline in vitro,they could showgolden-yellow fluorescence under the fluores-cence microscope,indicating the presence of newlyformed bone substances, of which both the quanti-ties and volumes were larger in R.S.M,groupthan those in the controls
