谷氨酸受体在大鼠c-foS基因的表达及其与癫痫易感性的关系 被引量:3

摘要 为探讨N一甲基一D一天冬氨酸(NMDA)受体在c一fos基因表达及其与癫痫易感性的关系,我们以Northern印迹杂交法测定结果显示:(1)外源性NMDA可诱导脑细胞c一fos基因的mRNA表达;竞争性或非竞争性NMDA受体拮抗剂可完中阻断c一fos基因mRNA表达。(2)NMDA诱导脑内c一fos的mRNA表达随神经元发育呈上升趋势,于体外培养24天达高岭。(3)在发育中,听源性癫痫易感大鼠脑细胞的c一fos基因mRNA表达均高于对照组,且在细胞培养18天时差异有显著意义。提示NMDA受体活性的变化可能与癫痫易感性的形成有关。 e use NMDA to induce expression of c-fosmRNA as a marker to observe the activity ofNMDA receptor in neurons during development, andcompare the activity of NMDA receptor betweenaudiogenic epilepsy -prone (P77PMC) andaudiogenic epilepsy resistant rats brain. In primaryculture of rats cerebral cortical neurons NMDA in-duced c-fos mRNA expression exhibites dose andtime-dependent changes, which can be prevented byantagonists. During the development of neurons, theNMDA -induced c-fos mRNA expression reaches amaximum at day 24. NMDA-induced c-fos mRNAexpression of P77PMC rats is higher than that of con-trols during 6 to 24 days in vitro with significant difference (P<0.05) at day 18. To present changes incfos mRNA expression induced by NMDA in cul-tured P77PMC rat cortical neurons may be one ofthe factors related to susceptibility of epilepsy inP77PMC rats
