p53蛋白异常表达与乳腺癌淋巴结转移相关性研究

THE RELATION BETWEEN P53 EXPRESSION AND LYMMPH NODE METASTASIS OF BREAST CANCER

应用抗ｐ５３单克隆抗体及快速免疫组化染色法检测了５０例石蜡包埋乳腺癌及７例乳腺良性增生性疾病ｐ５３蛋白的表达，结果显示１２例（２４％）乳腺癌呈阳性反应，７例良性病变均为阴性、在一组原发及淋巴结转移乳腺癌配对标本中，同一病人两个部位ｐ５３表达差异无明显意义，提示ｐ５３基因异常发生在肿瘤转移之前。在与临床病理指标的相关分析中发现，ｐ５３蛋白异常表达与乳腺癌淋巴结转移呈非常显著的相关关系（Ｐ＜０．００５），与髓样癌亦显著相关，４例髓样癌中，２例有淋巴结转移，ｐ５３蛋白强阳性。结果表明：ｐ５３蛋白不仅与乳腺癌发生有关，而且在乳腺癌淋巴结转移中起重要作用，因此，检测ｐ５３蛋白的表达可作为临床预测乳腺癌淋巴结转移趋势的重要参考指标之一。

Abstract The monoclonal antibody to p53 and a rapid immunohistochemical method were used in thisstudy to detect p53 expression in 50 paraffin-embedded breast cancers and 7 benign hyperplasticdiseases of breast, The results of immunostaining showed that 1 2 (24%) cases of breast cancers werepositive and all benign lesions were negative. No significant difference was found between primaryand metastatic sites of 11 paired samples of breast cancrs in p53 exprecssion, suggesting that the ab-normal expression of had p53 occurred before lymph node metastasis of breast cancer. Statistical analysisrevealed that the abnorrnal expression of p53 was significantly correlated with lymph nodemetastasis of breast cancer(P<0.005) and also correlated with medullary breast cancer in which twoof four cases strongly positive to p53 had lymph node metastasis. Our results indicate that the abnormal expression of p53 is not only involved in thedevelopment,but also involved in the metastasis of breast cancer. Therefore, detection of p53 expres-sion may have significant clinical lmportance in the evaluation of the tendency of lymphaticmetiastasis of breast cancer .
