肾癌相关蛋白G250 mRNA在肾细胞癌中表达的研究

Semi-quantitative reverse transcription-PCR assay for renal cell carcinoma-associated protein G250 mRNA

目的探讨肾癌相关蛋白G250mRNA在肾细胞癌(RCC)中的表达及其临床意义。方法采用RTPCR方法检测7例正常肾组织和33例RCC组织中G250mRNA的表达。33例RCC中透明细胞癌28例,非透明细胞癌5例;T18例、T213例、T312例;G111例、G215例、G37例。结果33例RCC组织中G250mRNA表达阳性28例(84.8%),7例正常组织G250表达均为阴性,2组间差异有统计学意义(P<0.01)。肾透明细胞癌组织G250mRNA表达率高于非透明细胞癌[96.4%(27/28)和20.0%(1/5),P<0.05]。T1、T2、T3RCCG250mRNA表达量分别为0.32±0.21、0.41±0.16、0.38±0.14,表达量与临床分期无相关性(P>0.05)。G1、G2、G3RCCG250mRNA表达量分别为0.48±0.32、0.23±0.16、0.11±0.11,表达量与病理分级呈负相关(P<0.05)。结论G250mRNA表达与RCC恶性程度有相关性,有望成为RCC诊断及预后判断的指标。

Objective To evaluate the expression of G250 mRNA in patients with renal cell carcinoma (RCC) and its clinical significance. Methods A semi-quantitative reverse transcription (RT)-PCR was developed to assay the levels of G250 mRNA obtained from 7 normal renal tissues and 33 cases of RCC.Of the 33 RCC cases,28 had clear cell carcinoma and 5 had non-clear cell carcinoma.By UICC staging, 8 cases were diagnosed with T_1 stage carcinoma,13 with T_2 and 12 with T_3.Pathological grading showed 11 cases of G_1,15 of G_2,and 7 of G_3. Results Among the 33 cases of RCC,G250 mRNA expression was present in 28 cases (84.8%).However, it was absent in all the 7 normal renal tissues.There was a significant difference between the 2 groups (P<0.01).The G250 mRNA expression rate in renal clear cell carcinoma [96.4% (27/28)] was higher than that in non-clear cell carcinoma [20.0% (1/5),P<0.05].The G250 mRNA expression levels in RCC cases of T_1,T_2 and T_3 were 0.32±0.21,0.41±0.16 and 0.38± 0.14 ,respectively (P>0.05);while they were 0.48±0.32,0.23±0.16 and 0.11±0.11,respectively,in RCC cases of G_1,G_2 and G_3,which showed that the expression of G250 mRNA was inversely correlated with tumor grades(P<0.05). Conclusions The G250 mRNA expression has correlation with malignancy of renal cell carcinoma, therefore it may serve as a useful marker for the diagnosis and evaluation of prognosis in patients with RCC.
