摘要 目的:观察神经干细胞(NSCs)植入AD大鼠海马内的存活情况,以及对学习记忆能力的影响。方法:采用神经球连续形成法从新生大鼠海马齿状回分离、培养NSCs,并用Nestin细胞免疫化学方法进行鉴定。NSCs经Hoechst33258标记后植入Aβ1-40诱导的AD模型大鼠海马,4周后,行Y迷宫实验检测大鼠的学习记忆能力,然后取脑进行荧光观察。以正常大鼠和AD模型大鼠为对照。结果:从新生大鼠海马齿状回成功分离培养了NSCs。NSCs移植组大鼠的学习能力和记忆能力较AD大鼠有明显提高。移植的NSCs能在海马存活并沿海马回迁移。结论:NSCs移植能改善AD模型大鼠的学习和记忆能力。 Aim: To investigate whether transplantation of neural stem cells can ameliorate learning and memory abilities of Alzheimer' s disease (AD)rats. Methods: Neural stem cells were isolated from dentate gyrus of hippocampus of newborn rats and were identified by nestin immunocytochemistry. After labeled with Hoechst33258, neural stem cells were grafted into the hippocampus of AD rats. Four weeks after transplantation, the rats underwent behavioral test using Y-maze trial. Fluorescence observation was used after the rats were sacrificed. AD rats and normal rats were used as control.Results: Neural stem cells from dentate gyrus of hippocampus of newborn rats had been successfully cultured and staining shown the nestin positive. Compared with AD rats,the learning and memorizing ability of rats in transplatation group was improved.Survival and migration of neural stem cells were demonstrated in the rats. Conclusion: Transplantation of neural stem cells can ameliorate memory deficits in AD rats.
