染氟大鼠生精细胞端粒酶逆转录酶表达的测定 被引量:2

摘要 目的:探讨氟对大鼠生精细胞端粒酶表达的影响。方法:30只雄性SD大鼠随机等分为对照组、高剂量染毒组、低剂量染毒组,分别腹腔注射蒸馏水、20mg/kgNaF和10mg/kgNaF溶液39d后,采用放射免疫法检测血清中雌二醇水平,原位杂交法检测端粒酶逆转录酶(TERT)表达情况,同时观察成熟精子质量和睾丸病理学的变化情况。结果:低剂量染毒组、高剂量染毒组雌二醇水平、TERT表达水平、精子计数和活动率均低于对照组(P均<0.05),精子畸形率高于对照组(P<0.05)。高剂量染毒组精子计数、精子畸形率和活动率与低剂量染毒组比较,差异均有统计学意义(P均<0.05)。结论:氟可降低TERT表达水平从而对生殖系统造成损害。 Aim: To study the effect of fluorine on telomerase expression of germ cells in testes of rats. Methods: A total of 30 male SD rats were randomly divided into control group,high GAAB2 and low-dose fluorine treated groups, which were given normal saline,20 mg/kg sodium fluoride,and 10 mg/kg sodium fluoride,respectively.The changes of estrogen and telomerase reverse transcriptase(TERT) were observed using radioimmunoassay and in situ hybridization, respectively.The changes of spermatozoon quality and pathological alterations were also observed.Results: The level of estrogen,the expression of TERT,and the number of the spermatozoon in high-and low-dose fluorine groups were lower than those of control group( P< 0.05).The deformity rate of spermatozoon in high-and low-dose fluorine groups was higher than that in control group( P< 0.05).Conclusion: Fluorine possibly damages the male reproductive system by reducing the expression of TERT.
