神经节苷脂(GM-1)对体外培养SH-SY5Y细胞兴奋性氨基酸毒性损伤的作用 被引量:12

摘要 目的探讨神经节苷脂(GM1)对体外培养SH SY5Y细胞兴奋性氨基酸毒性损伤的保护作用。方法采用细胞培养法,以神经母细胞瘤SH SY5Y细胞系为材料,制备兴奋性氨基酸Glu毒性损伤的离体细胞损伤模型。通过细胞形态学观察、MTT法测定细胞存活率观察不同浓度及不同时间GM1对上述损伤细胞的保护及治疗作用。结果GM1可增强SH SY5Y细胞的存活,抑制兴奋性氨基酸对细胞的损伤。与损伤组相比GM1治疗组均好于损伤组,且GM1高浓度组好于低浓度组。GM1对损伤SH SY5Y细胞MTT代谢率的影响显示,相同浓度GM1作用不同时间相比较12h优于6h,24h与12h无明显差异。结论GM1可促进神经细胞生存,对神经细胞具有明显保护作用。 Objective Investigate the protective effect of GM-1 to impaired SH-SY5Y cultured in vitro by (excitatory) amino acids. Methods Prepare the SH-SY5Y cellular damage model in vitro of excitatory amino acids. And observe the protective effect of GM-1 by different methods. Results GM-1 can improve the survival rate of SH-SY5Y, and inhibit the damage of excitatory amino acids. Treatment of GM-1 can protect SH-SY5Y with concentration-dependent and time-dependent character. Conclusions GM-1 can improve the survival rate of cell and protect neural cells.
