更年春方对卵巢切除大鼠骨与子宫组织雌激素受体亚型的调节 被引量:5

Regulation of Gengnianchun Formula on the Estrogen Receptor Subtype of Bone and Uterus in OVX SD Rats

摘要 目的探讨更年春方对卵巢切除大鼠骨与子宫组织雌激素受体(estrogen receptor,ER)亚型的调节.方法10~12月龄SD雌性大鼠卵巢切除建立骨质疏松模型.分别用更年春方、尼尔雌醇灌药4个月,RT-PCR法检测胫骨骨骺与子宫组织ER亚型mRNA的表达.结果大鼠卵巢切除后,血E2水平、椎骨骨密度、子宫湿重及胫骨骨骺与子宫组织ERα、ERβmRNA的表达均明显下降.尼尔雌醇治疗后椎骨骨密度、子宫湿重及胫骨骨骺与子宫组织ERα、ERβmRNA的表达均增高;更年春方组椎骨骨密度及胫骨骨骺与子宫组织ERβmRNA的表达增高,而子宫湿重及胫骨骨骺与子宫组织ERα mRNA的表达无明显改变.结论更年春方对卵巢切除骨质疏松大鼠有防治作用,对子宫无不良反应.其机制可能与其选择性上调胫骨骨骺与子宫组织ERβmRNA的表达有关. Purpose To evaluate the regulation of Gengnianchun formula (GNC) on the estrogen receptor (ER) subtype of bone and uterus in ovariectomized (OVX) SD rats. Methods Ten to Twelve months old SD rats were ovariectomized bilaterally to build the postmenopausal osteoporosis model.These rats were fed with GNC and nilestriol for 4 months,respectively.The semi-quantitative RT-PCR was performed to determine the mRNA level of ER subtype of tibia and uterus. Results Serum estridiol level,bone mineral density(BMD) of vertebra,wet weight of uterus and ERα,ERβ mRNA level of of tibia and uterus were reduced markedly after OVX.After treatment with nilestriol,BMD of vertebra,wet weight of uterus and ERα,ERβ mRNA level of tibia and uterus were increased dramaticly.GNC could improve BMD of vertebra and ERβ mRNA level of tibia and uterus,but had no action on the wet weight of uterus and ERα mRNA level of tibia and uterus. Conclusions GNC can improve BMD of vertebra of the OVX SD rats,without side effects on the uterus.The mechanism lies possibly in increasing ERβ mRNA level of tibia and uterus selectively.
