银杏天宝对大鼠实验性溃疡性结肠炎IFN-γ、IL-4的影响 被引量:5

Effects of Ginkgo Biloba Extract to the Level of IFN-γand IL-4 in Experimental Ulcerative Colitis in Rats

摘要 目的观察银杏天宝对实验性溃疡性结肠炎大鼠血清、大肠粘膜组织中IFN-γ、IL-4的影响。方法采用三硝基苯磺酸(TNBS)灌肠制备大鼠溃疡性结肠炎模型。将动物随机分为空白对照组、三硝基苯磺酸组、三硝基苯磺酸+生理盐水组和三硝基苯磺酸+银杏天宝组,观察肠道大体形态和组织学改变。采用ELISA法测定血清、大肠粘膜中的干扰素γ(IFN-γ)和白介素4(IL-4)含量。结果与三硝基苯磺酸组比较,三硝基苯磺酸+银杏天宝组损伤指数明显下降(P<0.01),大肠粘膜和血清IFN-γ浓度明显下降(P<0.01),IL-4浓度明显升高(P<0.01),IFN-γ/IL-4比值明显下降(P<0.01)。结论银杏天宝对溃疡性结肠炎有保护作用,抑制IFN-γ、提高IL-4生成可能是其作用机制之一。 Objective To investigate the effects of Ginkgo Bilobe extract(EGb) on the level of IFN-γ and IL-4 in serum and colonic mucosa of rats with experimental ulcerative colitis. Methods The rat ulcerative colitis model was established by using 2.4.6-trinitrobenzene sulfonic acid(TNBS).All rats were divided into four groups:normal group TNBS group ,TNBS+NS group and TNBS+EGb group.The macroscopical and histological changes of the colon were evaluated.The IFN-γ and IL-4 produced by serum and colonic mulosal was analyzed with ELISA. Results In TNBS+EGb group,the macroscopical and histological score was significantly lower than that of TNBS group( P <0.01).In the TNBS+EGb group,a lower level of IFN-γ production serum and colonic mucosa( P <0.01) was noticed in comparison with TNBS group.Compared with TNBS group,significantly increased IL-4 was noticed( P <0.01) in TNBS+EGb group. The ration of IFN-γ/IL-4 was lower in TNBS+EGb group compared with TNBS group( P <0.01).Conclusion Ginkgo Biloba extract has protective effect on ulcerative colitis of rat and this effect may be caused by lowering the level of IFN-γ and raising the level of IL-4 in serum and colonic mucosa.
