摘要
以血清Ⅰ型鸭疫里默氏菌(RA)分离株为菌种研制RA油佐剂灭活疫苗,对3日龄樱桃谷鸭颈背皮下注射0.25mL/只即可产生良好的免疫应答,免疫后10、13和16天对同源RA的攻击表现出35%、85%和100%免疫保护。疫苗一次免疫后30天ELISA抗体滴度达到高峰,至93日龄时仍能检出抗体。10日龄进行第二次免疫可产生更高的抗体滴度,到65天时仍能保持较高抗体水平。ELISA抗体滴度≥500时雏鸭即获得免疫保护。疫苗免疫雏鸭后能够显著(P<0.05)或极显著(P<0.01)地增强T细胞的免疫力,时间持续两周。RA油佐剂灭活疫苗免疫雏鸭后免疫保护的形成是体液免疫与细胞免疫协同作用的结果,其中体液免疫占主导作用。
Riemerella anatipestifer(RA) inactivated vaccine in oil adjuvant was made with serotypes Ⅰ isolated strains.Excellent immunity could be got when 3-day-old cherry valley ducklings were inoculated subcutaneously with a dose of 0.25 ml. When 10, 13 and 16 days post inoculation(PI), 35%, 85%and 100% vaccined ducklings can be against the challenge of homologous RA respectively. Indirect Enzyme-linked Immunosorbent Assay (ELISA) showed that specific antibody peaked 30 days PI in titers and could still be detected 93 days PI. Antibody titers in ducklings inoculated two times (3 and 10-day-old ) was higher than the ducklings inoculated one time in 3-day-old, and was still high 65 days PI. When ELISA titers ≥500, the vaccined ducklings could have good immunity. The notably enhanced cell mediated immunity in the experiment group was different (P<0.05) or very different (P<0.01) from the control and was about 2 weeks in duration.The vaccine attained its efficacy by cooperation of humoral immunity and cell mediated immunity it stimulated, and humoral immunity had dominant function.
