摘要 以粳稻"日本晴"成熟种子诱导愈伤组织,统计愈伤诱导率及绿苗分化率,分析培养基中2,4D和6BA的不同浓度配比对培养力的影响,得到二者的最适配比,愈伤组织的质量和组织培养中再生植株的频率都有较大提高.实验结果表明:2,4D是诱导愈伤组织必不可少的因子,与6BA配合使用对愈伤组织的培养效果更好.对"日本晴"而言,当2,4D为2.0mg/L,6BA为0.5mg/L时愈伤组织的诱导率最高;当2,4D为2.0mg/L,6BA为0.2mg/L时,绿苗分化率和培养力最高. In this experiment,Oryza sativa ssp.japonica Nippobare mature seeds were used to initiate calli and NB media supplemented with different concentrations and combinations of 2,4D (2,4dichlorophenoxy acetic acid) and 6BA(6benzylaminopurine)was used for callus initiation.The effects of the two plant horome were analyzed by counting the initiation rate and regeneration rate.The results showed that 2,4D was a necessary factor in callus initiation,the media with both 2,4D and 6BA would have a better effect on mature embryo culture.For Oryza sativa ssp.japonica Nippobare 2.0?mg/L of 2,4D and 0.5?mg/L of 6BA added in culture media express the highest initiation rate;2.0?mg/L of (2,4D) and 0.2?mg/L of 6BA added in culture media express the highest regeneration rate and culture ability.
