摘要
从患病雏鹅的病料样本中分离鉴定出一株小鹅瘟野生强毒。采用鹅胚增殖病毒,以琼脂扩散试验从感染鹅胚尿囊液中检出小鹅瘟病毒抗原;以感染鹅胚尿囊液人工接种5日龄健康雏鹅,复制出与自然感染病例相类似的病变;通过电镜观察到典型的小鹅瘟病毒粒子。该分离毒株核酸分子量为5000bp。制备的小鹅瘟沉淀抗原可以检测患病雏鹅血清中的抗体,监测治疗血清的滴度和疫苗接种的效果。标记的小鹅瘟荧光抗体能直接检测患病雏鹅组织中的病毒抗原。针对编码主要结构蛋白VP3的基因设计合成一对引物,扩增出与预期长度相符的特异性DNA片段。
A virulent virus of gosling plague virus(GPV)was isolated and identified from affected goslings.The viruses were propagated in goose embryos,the GPV antigen could be detected from allantoic fluid of infected embryos by agar gel precipitation.At the age of 5 days, the goslings were artificially inoculated with virus sample and the pathological lesions were similar to the natural infected cases. The typical gosling plague virus was observed by electronic microscope.The molecular weight of nucleic acid was determined as 5 000 base pairs. The prepared antigen of GPV could detect the serological antibodies in affected goslings, survey the titers of therapeutic sera on market and investigate the effects of vaccine immunization. The fluorescent antibody of GPV could detect the virus antigen in tissues of affected goslings.A pair of primers were designed and synthesized, which were directed to the gene coded for VP3.The amplified DNA fragment was in accordance with the expected length.
