摘要
研究了乌塌菜(Brassia narinosaBailey)优良变异植株叶片愈伤组织的诱导、分化以及试管苗生根、移栽、扦插及田间栽培所需要的条件,并建立起乌塌菜优良变异植株的无性系及快速繁殖技术.结果证明,诱导乌塌菜愈伤组织的理想培养基是MS+BA 2.0 mg/L;诱导愈伤组织分化的理想培养基是MS+La(NO3)31.0 mg/L+BA 0.5 mg/L;生根的理想培养基是12MS+IAA 1.0 mg/L;以炉灰渣为试管苗的移栽扦插基质,移栽成活率为93%,扦插成活率为88%.耐寒乌塌菜试管苗的田间栽培试验还证明,这种试管苗生长旺盛,根系发达,产量可提高10%.
Coldresistant mutation clone of Brassia narinosa Bailey is used as the researching subject.The conditions needed in callus induction,differentiation,tube spout radication,transplantation and graftage of Brassia narinosa Bailey have been studied,also its regenerative clone and rapid propagation technique are established.The best medium for callus induction is MS+BA 2 mg/L.The high frequency of differentiation is obtained in MS+La(NO_3)_3 1 mg/L+ BA 0.5 mg/L.12MS+IAA 1 mg/L is suitable for radication.With cinder used as seeding matrixs,the survival percent of transplantation could reach 93% and that of graftage is 88%.Field planting showed that the tube spout grew well with roots flourishing and the yield increased by 10%.
