HPLC-ELSD法测定补中益气片中黄芪甲苷的含量

Determination of Astragaloside IV in Buzhong Yiqi Pian by HPLC-ELSD

目的建立补中益气片中黄芪甲苷含量的测定方法。方法采用HPLC-ELSD蒸发光散射检测法,色谱柱为ODSC18(4.6mm×250mm,20μm);流动相:乙腈-水(1∶2);进样10μL;室温;流速:0.8mL/min;漂移管温度:105℃;气体流速:2.70mL/min。结果黄芪甲苷对照品在2.528～12.640μg范围内,进样量与峰面积呈良好的线性关系,r=0.9995,平均回收率为99.25%。结论本方法操作简便,结果准确,重现性好,可用于控制该制剂的质量。

Objective To establish a method to determine Astragaloside IV in Buzhong Yiqi pian. Methods HPLC-ELSD was used to determine directly Astragaloside IV on ODS C18 column (4.6 mm×250 mm, 20 μm) (Echrom Dalian). Using CH3CN-H2O (1∶2) as a mobile phase, injecting amount 10 μL, 25 ℃, the flow rate was 0.8 mL/min. The temperature of drift tube for ELSD was 105 ℃ and the flow rate of N2 was 2.70 mL/min. Results Astragaloside IV showed good linear relationship in range of 2.528～12.640 μg, r =0.9995, the average recovery was 99.25%. Conclusion The method is simple, accurate, practical, and can be used for the determination of this preparation.
