摘要
试验研究了蝴蝶兰'红日'花梗离体培养启动分化的光照强度和继代增殖培养的容器类型及培养基配方.结果表明,以H+6-BA 12.0 mg/L+NAA 0.4 mg/L+IBA 0.4 mg/L +CM 100 mL/L+柠檬酸50 mg/L作启动分化培养基,光照强度在300~500 lx较好;继代培养用PP,PC透明塑料容器与玻璃容器对原球茎的增殖、芽的分化效果相近;6-BA低浓度促进分化芽,高浓度促进分化原球茎;以H+6-BA 0.3 mg/L+NAA 0.2 mg/L+CM 100 mL/L的培养基对继代和壮苗为好,椰子汁对芽分化和壮苗有促进的作用.
The effect of illumination intensities, types of vessel and media used as propagation on polarization of flower-stems of Phalaenopsis “Redsun” in vitro culture was studies. The results showed that H+ 6-BA 12.0 mg/L+NAA 0.4 mg/L+IBA 0.4 mg/L+CM 100 mL/L+citric acid 50 mg/L was suitable for pedicel polarization under the illumination of 300~500 lx; the effect of PP, PC plastic vessels on the propagation of protocorm-like body(PLB) and bud polarization were near to glass vessels; low and high concentration 6-BA promoted the polarization of bud and PLB respectively; H+6-BA 0.3 mg/L+NAA 0.2 mg/L+CM 100 mL/L was good medium for culturing and strengthening seedlings, coconut milk could make the buds stronger.
