摘要
目的:探讨并研究磁性纳米微粒对不同种生物样品中蛋白质的分离方法及效果。方法:磁性纳米微粒与AEAPS反应后,表面修饰并连接-NH2制成蛋白质分离器,通过氨基与羧基的结合及外磁力作用分离蛋白质。结果:MNP-NH2对BSA的结合呈现直线相关(r=0.9942,p<0.001)。对几种来源不同的生物样品中可溶性蛋白的分离结果显示,各实验组数值与对照组比较,均具有显著性差异(P<0.05~0.001)。结论:MNP-NH2对于分离生物样品中蛋白质具有较好的分离效果对蛋白质活性影响较小。
Objective: The wish of the work is to study the effect of protein selection from several traditional Chinese medicine with MNP-NH2. Method: As a result of the reaction to AEAPS,magnetic nanoparticle was modified and joined -NH2 on its surface. This material was called MNP-NH2 as a whole. MNP-NH2 can be used in selection of protein. Results: The combine between MNP-NH2 and BSA has a linear correlation(r=0.9942,p<0.001). Forethmore,several proteins from different traditional Chinese medicines were selected by MNP-NH2. Compared with the control groups,the results of experiment groups have significant difference(P<0.05--0.001). Conclution: This research indicated that MNP-NH_2 can be extensively used in protein selection.Meanwhile,this is a effective new way to realized the modernization of traditional Chinese medicine.
