摘要 以珂字312和冀无2031为材料,对陆地棉体细胞胚胎发生技术进行了改进,建立了一套培养程序。在悬浮培养中,加入50g·L-1的PEG6000的液体培养基可提高悬浮培养后正常胚的萌发率,对胚状体萌发有促进作用。悬浮培养后的胚性愈伤组织继代到含有80-160mg·L-1PEG6000的胚性愈伤组织萌发培养基中,暗培养1-2周,胚性愈伤组织明显疏松;在光照条件下继续培养两周可形成大量绿色萌发胚。加入500mg·L-1的CH及NH4NO3减半、KNO3加倍的培养基利于胚状体的分化,且异常胚的比例低。 Two varieties of upland cotton, Coker 312 and Jiwu 2031, were studied on somatic embryo-genesis. They produced different amount of embryogenic calli and regenerated plants. A better procedure of cotton embryogenic callus inducement was established. Special effects of PEG6000 on somatic embryogenesis in upland cotton were investigated. PEG6000 could raise ratio of embryoids germination remarkablely, which got the highest rate of embryoids germination in the treatments of 50 g·L-1 in suspension culture. It promoted embryogenic callus germination in normal light period in two weeks after one or two weeks in darkness with 80-160 mg·L-1 of PEG6000 in solid medium. This method was also applied to maintenance embryonic callus. MSB(MS inorganic salts and B5 vitamin) with phy-tohormone free, 500 mg·L-1 of CH, half amount of NH4NO3 and double amount of KNO3 could stimulate formation and germination of embryoids, and reduce abnormal embryoids.
