摘要
目的检测强直性脊柱炎(AS)患者关节积液与外周血树突状细胞(DC)的数量变化及成熟度。方法利用组合性荧光抗体(CD3,CD14,CD16,CD19,CD20,CD56)设门从白细胞中选定树突状细胞,以30例健康人作对照,用流式细胞仪检测30例AS患者病变关节积液和外周血CD123+、CD11c+的树突状细胞的数量。结果AS静脉血CD11c+DC在白细胞中所占比例为(0.30±0.09)%,比健康人静脉血中CD11c+DC所占比例(0.18±0.08)%,高(t=5.459,P<0.001);AS关节积液CD11c+DC在白细胞中所占比例(0.45±0.11)%,比AS静脉血CD11c+DC在白细胞中所占比例高(t=5.781,P<0.001);AS静脉血CD123+DC在白细胞所占比例(0.15±0.08)%,与健康人静脉血CD123+DC所占比例(0.12±0.07)%无显著差异(t=1.546,P>0.05);AS关节积液CD123+DC在白细胞中所占比例(0.23±0.08)%,比AS静脉血CD123+DC在白细胞中所占比例高(t=3.874,P<0.001)。结论AS患者病变关节局部高、低成熟度的DC数量均有明显增多,而静脉血中主要表现为高成熟度DC增多,这可能反映AS患者尤其病变局部抗原提呈水平显著高于健康人。DC与AS间存在密切的联系。
Objective To research the amount and maturity of dendritic cell (DC) in joint fluid and peripheral blood of ankylosing spondylitis (AS) patients.Methods The dendritic cells from white blood cells (WBC) were selected by setting up the gate in flow cytometer with combined fluorescence antibodied (CD3,CD14,CD16,CD19,CD20,CD56).The amount of CD11c + DC and CD 123 + DC in joint fluid as well as peripheral blood of ankylosing spondylitis patient (n=30) and normal healthy subjects (n=30) were measured simultaneously by flow cytometer.Results The percentage of CD11c+ DC in peripheral blood of AS patients(0.30±0.09%) was more than that in control group (0.18±0.08%) ( t =5.459, P <0.001).The percentage of CD11c+ DC in joint fluid of AS patients(0.45 ±0.11%) was more than that in peripheral blood( t =5.781, P <0.001).There was no difference of percentage of CD123+ DC in peripheral blood between AS patients(0.15 ±0.08%) and control group (0.12±0.07%) ( t =1.546, P >0.05).The percentage of CD123+ DC in joint fluid of AS patients (0.23±0.08%) was more than that in peripheral blood( t =3.874, P < 0.001 ).Conclusions Increased DC amount with both high and low maturity was found in joint fluid of AS patients, while only increased DC amount with high maturity was found in peripheral blood of AS patients.Thus,a high level of antigen presentation in AS patients is exhibited and there is a close relationship between DC amount and AS.
