摘要
目的为寻找适合临床应用的新型MDR逆转剂,观察榄香烯乳注射液对人胃癌耐药细胞系SGC7901/VCR的影响作用。方法采用MTT法进行药敏实验,并通过流式细胞仪检测细胞周期变化、胞浆药物浓度,免疫细胞化学染色法显示耐药细胞中MRP和P-gp蛋白的分布。结果榄香烯对SGC7901/VCR的半数抑制浓度为18.65μg/mL,并且能够逆转其对VCR、ADM的耐药性,但对5-FU、DDP的耐药性无明显影响。经榄香烯诱导后细胞内药物浓度明显增加,S期和G2+M期增加,G1期减少,MRP和P-gp的表达明显减弱。结论榄香烯能够逆转SGC7901/VCR的耐药性,其机理与促使MRP和P-gp蛋白表达减少有关,有待于进一步研究。
Objective To observe the reversion effect of elemene on human drug-resistant gastric cancer cell line SGC_ 7901 /VCR. Methods The cell cycle alteration,intracellular drug concentration of these cells,the sensitivity of gastric cancer cells to several anti-tumor drugs and expression of P-gp and MRP in gastric cancer cells were detected by FCM,MTT assay and immunocytochemical staining,respectively.Results The IC50 value of elemene to SGC_ 7901 /VCR was 18.65μg/mL. Elemene could reserve the multidrug resistance(MDR) of SGC_ 7901 /VCR to VCR and ADM.but SGC_ 7901 /VCR sensitivity to 5-FU and DDP had no obvious change.By comparing with SGC_ 7901 /VCR, SGC_ 7901 /VCR with P-gp and MRP cells decrease in the G1 stage and increase in the S and G2+M stage. P-gp and MRP expression level was lower than that without elemene. Conclusion Elemene could reserve the multidrug resistance(MDR) of SGC_ 7901 /VCR,for which the decreased expression of P-gp and MRP maybe responsible.
