HPLC法测定伤湿外用酊中士的宁含量

Determination of Strychnine Continded in Shangshi Waiyong Tincture by HPLC

目的建立伤湿外用酊有毒成分士的宁含量测定方法。方法采用高效液相色谱法;以C18色谱柱为固定相;以正己烷-二氯甲烷-甲醇-浓氨试液(42.5∶42.5:5.0:0.35)为流动相;检测波长260nm。结果士的宁在4～20μg/mL范围内,线性关系良好(r=0.9992),平均加样回收率为98.16%;方法精密度RSD=1.71%(n=5)。结论该方法操作简便、准确,可作为该酊剂中有毒成分士的宁含量的控制方法。

Objective: To establish amethod for determining stryclinme in Shangshi waiyong tincture. Method: The experimental conditions in HPLC were EclipseXDB-C_ 18 colum (5μm,150mm×4.6mm).The mobile phase consists of N-hexane:Methylene-chloride:Methanol:Ammonia solution(42.5:42.5:5.0:0.35). The flow rate was 1.0mL/min.Detection was done spectrophoto metrically at 260nm. Result: The brucing calibration curve was in good ltincanity. over the range of 4～20μg/mL(r=0.9992), average recovery was 98.16%, RSD=1.71%(n=5). Conclusion:This HPLC is simple rapid,accurate and issuitable for the quality control of Shangshi Waiyong tincture.
