摘要
目的探讨前列腺增生(BPH)组织中转化生长因子β1(TGFβ1)和碱性成纤维细胞生长因子(bFGF)的表达以及β射线内照射对TGFβ1和bFGF表达的影响。方法应用免疫组织化学法检测9例正常前列腺(NP)、15例增生前列腺及35例实施90Sr/90Yβ射线照射后前列腺增生组织中TGFβ1和bFGF的表达。结果NP上皮和间质细胞中TGFβ1阳性细胞率分别为68.2%±10.5%,29.7%±4.6%;BPH上皮和间质细胞中TGFβ1阳性细胞率分别为64.8%±9.3%,28.6%±4.1%。应用90Sr/90Yβ射线内照射后4、7、15d后前列腺上皮和间质中TGFβ1阳性细胞率明显升高,差异有显著性意义(P<0.01)。NP上皮和间质细胞中bFGF阳性细胞率分别为17.4%±3.7%,42.5%±6.8%;BPH上皮和间质细胞中bFGF阳性细胞率分别为46.3%±8.2%,73.2%±12.1%。应用90Sr/90Yβ射线内照射后bFGF阳性细胞率在上皮和间质细胞中明显下降(P<0.01)。结论TGFβ1和bFGF与前列腺细胞凋亡的调节有关,β射线能使前列腺组织中TGFβ1和bFGF含量发生变化,促使前列腺细胞凋亡。
Objective To investigate the expressions of TGF-β_1 and bFGF in benign prostatic hyperplasia (BPH) and the effect of β-rays on the expressions of TGF-β_1 and bFGF. Methods The expressions of TGF-β_1 and bFGF were detected by immunohistochemical method in 9 normal prostatic (NP) tissues and 15 hyperplastic prostatic tissues and 35 hyperplastic prostatic tissues treated with 90Sr/ 90Y β radiation. Results The positive rate of TGF-β_1 in epithelia and stroma of NP tissues was 68.2% ±10.5% and 29.7% ± 4.6%, while it was 64.8%±9.3% and 28.6%±4.1% in BHP tissues. The expression of TGF-β1 in epithelia and stroma of BPH treated with 90Sr/ 90Y prostatic hyperplasia applicator increased significantly 4,7,15 day after radiation(P<0.01). The expression of bFGF in epithelia and stroma of NP tissues was 17.4%±3.7% and 42.5%±6.8%, while it was 46.3%±8.2% and 73.2%±12.1% in BPH tissues. The expression of bFGF in epithelia and stroma of BPH treated with 90Sr/ 90Y prostatic hyperplasia applicator decreased significantly after radiation(P<0.01). Conclusions TGF-β_1 and bFGF gene may play an important role in the regulation of prostatic apoptosis, and the treatment of β-rays can change the TGF-β_1 and bFGF content of BPH tissues and accelerate the apoptosis of prostatic cells.
