HPLC法测定夏天无胶囊中原阿片碱的含量

Determination of Protopine in Rhizoma Corydalis Decumbentis Capsule by HPLC

目的建立夏天无胶囊中原阿片碱的含量测定方法。方法采用HPLC法,色谱柱:HypersilODS柱(4.6×250mm,5μm);柱温40℃;进样量5μL;流动相:甲醇-0.1%三乙胺溶液(65∶35),流速:1.0mL/min;检测波长:290nm;理论板数按原阿片碱计算应不低于4000。结果原阿片碱在0.3168～1.5840μg范围内,进样量与峰面积积分值之间线性关系良好,r=0.9996。平均回收率为100.6%,RSD=2.68%。结论该方法简便准确,重现性好,可用于夏天无胶囊的质量控制。

Objective To develop a HPLC method for the determination of protopine in Rhizoma Corydalis Decumbentis Capsules. Methods A HPLC was performed on a Hypersil ODS column( 4.6× 250 mm, 5 μ m) . The mobile phase was methanol - 0.1 % water solution of triethylamine (65∶ 35). Determination wavelength was 290 nm and flow rate was 1.0 mL/min. The theoretical plates should over 4000 according to the content of protopine. Results A good linearity was shown in the range of 0.3168～ 1.5840 μ g, r=0.9996. The average recovery was 100.6 % , RSD=2.68 % . Conclusion The method is simple, effective and can be used for the quality control of Rhizoma Corydalis Decumbentis Capsules.
