摘要
目的:探讨Th1/Th2细胞在免疫性肝损伤小鼠中的作用。方法:应用尾静脉注射卡介苗(BCG)和脂多糖(LPS)建立免疫性肝损伤小鼠模型,分离小鼠淋巴细胞,经佛波醇乙酯(PMA)、离子霉素、莫能菌素刺激培养6小时后,收获细胞,利用细胞内细胞因子染色流式细胞术检测Th1/Th2比例;另外经刀豆蛋白A(ConA)刺激培养72h后,收集培养上清,ELISA法检测IL-2、IFN-γ、IL-4、IL-10的水平,同时观察地塞米松对Th1/Th2细胞的影响。结果:在模型组,随着时间的延长,小鼠外周血PBMC中Th1型细胞因子IL-2、IFN-γ水平逐渐升高,Th2型细胞因子IL-4、IL-10水平逐渐降低,Th1细胞的百分率逐渐升高,Th2细胞的百分率逐渐降低,Th1/Th2的比值逐渐增大,与肝组织损害程度呈现正相关。地塞米松能够抑制Th1型细胞因子的分泌,降低Th1细胞的百分率,恢复Th1/Th2的比例,对肝组织损害具有保护和改善作用(P<0.05)。结论:Th1/Th2细胞失衡是造成小鼠免疫性肝损伤的原因之一,Th1应答占主导地位。
Objective:To investigate the role of Th cells in the mouse with immunological liver injure.Methods:The ratios of Th_1/Th_2 cells in the mouse with immunological liver injure were detected by flow cytometry with two color intracellular cytokine staining;The levels of IL-2、IFN-γ、IL-4 and IL-10 were detected by ELISA.Results:In the model group,the levels of IL-2、IFN-γ were gradually higher,the levels of IL-4、IL-10 were gradually lower,Th_1 cell ratios were higher,Th_2 cells ratios were lower,the ratios of Th_1/Th_2 were higher,there was positive correlative between the ratio of Th_1/Th_2 and liver injure.Dexamethasone could downregulate the levels of IL-2、IFN-γ and the radio of Th_1 cell,and could convert the ratio of Th_1/Th_2,and could improve liver pathological injury(P<0.05).Conclusion:The imbalance of Th_1/Th_2 was one of the causes of mouse immunological liver injure,Th_1 cells were predominate.
