环氧化酶-2、血管内皮细胞生长因子、热休克蛋白27和P53在非小细胞肺癌中的表达及其临床意义 被引量:2

摘要 目的通过对中国人非小细胞肺癌(NSCLC)患者肿瘤组织中环氧化酶-2(COX-2)、血管内皮细胞生长因子(VEGF)、热休克蛋白27(HSP27)和P53表达的检测,观察这4种蛋白质在NSCLC肿瘤组织中的表达情况,分析其与性别、年龄、病理类型、肿瘤细胞分化程度、肿瘤直径、淋巴结转移的相关性,为临床判断和有效控制NSCLC提供有价值的参考。方法采用免疫组织化学法检测50例NSCLC患者肿瘤组织中COX-2、VEGF、HSP27和P53的表达,采用原位杂交法检测COX-2mRNA的表达。结果COX-2、VEGF、HSP27、P53单一指标阳性均与性别、年龄、组织学分类、肿瘤直径、N分期、肿瘤细胞分化程度无关。COX-2与淋巴结转移有关(偏相关系数为0.3237,P=0.028),而VEGF、HSP27、P53与淋巴结转移无关。COX-2和P53或COX-2和VEGF共同高表达在N2期的NSCLC中明显多于N0和N1期;COX-2、P53和VEGF共同高表达或其中两种蛋白质高表达在N2期的NSCLC中明显多于N0和N1期(P=0.002)。HSP27高表达虽然与淋巴结转移无关,但与COX-2高表达有关(偏相关系数为0.5143)。结论COX-2、P53和VEGF共同高表达或其中两种蛋白质高表达与NSCLC淋巴结转移密切相关。COX-2与NSCLC淋巴结转移有关。 Objective To investigate the relevance of cyclooxygenase-2 (COX-2), vascular endothelial growth factor (VEGF), heart shock protein 27(HSP27), P53 and clinical features of non-small cell lung carcinoma (NSCLC) for raising the diagnostic accurracy and effective control. Methods The detection of COX-2,VEGF,HSP27 and P53 from formalin-fixed paraffin-embedded lung cancer tissue of 50 Chinese NSCLC cases,were carried out by immunohistochemistry. COX-2 mRNA was detected by in situ hybridization. Results In those cases, the high expression of COX-2,VEGF,HSP27 or P53 singly had no relationship with sex, age, histological subtypes, differentiation, tumor diameter or lymph node metastasis. COX-2 expression was correlated with lymph node metastasis by partial correlation analysis. The coefficient was 0.3237. Coexpressions of COX-2 and P53 or COX-2 and VEGF were more in N2 than those in N1 and N0; coexpressions of COX-2,P53 and VEGF or two of the three were much more in N2 than those in N1 and N0 (P=0.002). Conclusions The coexpression of COX-2,P53 and VEGF or two of the three is being more in lymph node metastasis of Chinese patients NSCLC,COX-2 is relevant to NSCLC lymph node metastasis.
