摘要 目的探讨组织芯片技术高通量原位免疫组织化学检测石蜡包埋肝细胞癌组织的价值。方法1991年1月-2002年6月西南肝胆医院实施根治性肝切除,病理证实为肝细胞癌234例石蜡组织块,健康成人肝组织18例(西南医院肝胆外科研究所肝移植供肝组织)作为对照,重新包埋石蜡组织,制备成为2个微阵列,分别为13×10(129点)和14×9(123点),微阵列点直径1.0mm,点间距为0.5mm。Envision法检测Vimentin和E cadherin的表达。结果组织芯片中组织样本的组织学结构完整,可满足病理学实验要求。结论组织芯片技术提供了一种快速、有效的组织学方法,可广泛应用大样本肝细胞癌的免疫组织化学检测。 Objective To explore the application and values of paraffin-embedded hepatocellular carcinoma (HCC) tissue microarray technique for high-throughput in situ immunohistochemical studies. Methods Paraffin-embedded tissue from 234 HCC and 18 healthy liver tissue samples from January 1991 to June 2002 at Southwest Hospital were used for the microarray. Two TMA blocks were constructed. 129 specimens were arrayed on one block and 123 on the other. Core tissue biopsies of (1.0) mm in diameter were taken from carefully selected morphologically representative regions of individual paraffin-embedded samples (donor blocks) and precisely arrayed into a new recipient paraffin block using a custom built instrument. Immunohistochemistry Vimentin and E-cadhenrin expression in HCC was determined using immunohistochemistry technique. Results The histological structure of the specimens was complete in tissue microarray. It can meet the requirements of pathological experiments. Conclusions Tissue microarray provides a rapid and powerful histological method that may be used for high-throughput in situ studies and play an important role in tumor biology.
